| Beschreibung: | Leaf senescence involves massive changes in gene expression. These changes are preceded by a decline in functionality of the photosynthetic apparatus. It is hence likely that chloroplast derived signals (retrograde signalling) are involved in induction of senescence associated gene expression. We have isolated Whirly1 binding to the promoter of the HvS40 gene which is highly expressed during senescence of barley leaves. Whirly1 is a multifunctional protein which was shown to be located in plastids and the nucleus of the same cell (Grabowski et al. 2008, Plant Physiology 147: 1800-1804 ). As a dually located protein Whirly1 is an excellent candidate for transducing information on chloroplast functionality to the nucleus. To investigate the impact of Whirly1 on leaf senescence, expression of the gene was knocked down by RNAi. Barley lines with low level of the HvWhy1 protein showed a delay in senescence when they were grown at high light intensity. A model for the mechanism of Whirly1 mediated control of senescence by plastids is presented. |
